Lawrence Van Hook was baptized on June 23, 1723, at the Old Dutch Reformed Church in New York. He is the son of Aaron van Hoek and Katherine Balck. Lawrence was likely born in New Jersey, but apparently his parents took him back to New York to have him baptized.

He was named for his paternal grandfather. Lawrence grew to maturity in New Jersey. Lawrence first wife was Bridget Lloyd, daughter of Thomas and Bridget Lloyd (also spelled "Loyd").

Lawrence and Bridget were probably married about 1750 as she is mentioned by her maiden name in her father's will 27 January 1748 and their first child was baptized early in 1751. The baptisms of their first three children are in the Tennent Church Records, Monmouth County, New Jersey. Lawrence is recorded as supplying some of the boards to build the Old Tennent Meeting House in March of 1750.

Lawrence moved to North Carolina (originally Orange County) from New Jersey with his father Aaron and other relatives about 1755. Their third child, Elizabeth, was baptized in New Jersey in May of 1755 and Lawrence's father is found in North Carolina late in the summer of 1755. Lawrence's father died 5 years later in 1760 and Lawrence was one of the executors of his father's will.
