Amos Mann was born in Nicholas County, Kentucky where he lived his entire life. Like his father Peter Mann, Amos was a farmer and landowner. On January 23, 1838, Peter deeded 343 acres of land to his son Amos.

Amos was a member of the Old Settlers Organization that met every June in Carlisle, Kentucky. The members were those whose families had been the earliest settlers of Nicholas County.
