John Moles was baptised on 15th July 1868 at Hitchin in Hertfordshire. He was the fifth of ten children of a midwife called Fanny Moles, formerly Worbey, and her husband Thomas Moles, an agricultural labourer and mole and rat catcher. The family lived at a row of cottages called The Riddy, which stood in the fields to the south-east of the town.

In the late 1870s the family left The Riddy (which appears to have been demolished immediately after they left) and moved a short distance to Blackhorse Lane, which forms the boundary between St Ippolyts and Hitchin. From at least 1881 to 1901 the family lived on the south side of Blackhorse Lane, putting them in St Ippolyts parish. John's father Thomas died in 1895, aged 57, and was buried at St Ippolyts parish church.

John grew up to follow his father's profession, also becoming an agricultural labourer and rat catcher. He never married and appears to have lived with his mother until her death.

Between 1901 and 1911 the family moved to the north side of Blackhorse Lane, putting them in the town of Hitchin. John's mother Fanny died in 1922, aged 80, and was buried in Hitchin Cemetery. John only outlived his mother by four years, dying in 1926, aged 58. He was buried at Hitchin Cemetery alongside his mother.
