Six Mile Run is an inhabited place.

The village grew up around a Dutch Reformed Church established in 1710.

The village is named for a stream which flows through both modern Somerset County and modern Middlesex County. Also, in earlier times after the establishment of the modern border between Somerset County and Middlesex County, the unincorporated community was construed as extending into both counties.
