Mount Olive Cemetery is located South-East of the junction between Delaware Routes 48, 100 and 141. The South border of the Cemetery rests along Lancaster Pike (DE 48/100). A single lane (track) runs into the Cemetery and circles near the center of the grounds (OpenStreetMap, 2012).

The Cemetery holds the remains of people who had previously been interred in at a former cemetery located at the corner of Lancaster Avenue and Union Street in Wilmington, Delaware, St. Peter (sic) Cemetery (Delaware Public Archives, 2007).

Cemeteries nearby include Mount Zion Cemetery, Silverbrook Cemetery and Cathedral Cemetery, each South-East from Mount Olive Cemetery along Lancaster Pike; and Beth Emeth Cemetery, which abuts the south border of Silverbrook Cemetery.
