It was founded in 1799 on land donated by William Campbell when the county was formed and named for General Nathanael Greene. The site is southeast of Caney Station, an earlier settlement. The Greenville post office opened in 1801. The population in 1990 was 4,689.

Its county seat is Greenville. The Central City Micropolitan Statistical Area includes all of Muhlenberg County. Central City is the largest city in the county with a 2010 Census of 5,978 residents.
