Named in his father’s 1668 will.

He was freeman in Weymouth in 1647 and in 1650 bought land in Cohinasset. In 1662 he was one of the ten citizens of Weymouth granted land in Mendon. He was assigned a house and forty acres, and was probably there by Sep 1663. In 1670 he purchased forty acres from Moses Paine wast of Mendon Village and this was inherited by his heirs, who as late as 1729 drew lands in his right in what is now Uxbridge. He was a Captain in King Philip's War, and probably shared in the suffering when Mendon was burned by the Indians. He went back to Weymouth and then went to live in Scituate, where he lived near the road from Cohasset to Scituate, having sold most of his Mendon lands in 1697. At some point before his death, he remarried to Abigail Wright Sharp Clapp, who is named in his will. He was a Congregationalist deacon.

[From MD, p. 42:138, Plymouth Probate 1:309, docket 10270] Will of William Holbrook of Scituate, dated 9 Sep 1696, inventory taken 11 Jul 1699, proved 17 Jul 1699, mentions "well beloved wife" Abigail Holbrooke, giving her what "she brought with her at marriage", "I doe give and bequeath unto my daughter Mehitable Sprague the sum of twleve pounds in money to be payd to her by her husband out of the twenty pounds which he oweth to me by bill and the reaminder of sayd bill of twenty pounds her sayd husband Jonathan Sprague shall pay unto my executor." Will also names son Cornelius and his daughter Mehetabell Holbrooke, son William, daughters Elizabeth Buck, Hopestill Reed, Jane Balcom/Balcombe, and Jane's daughter Albee. "Eldest son Samuel Holbrooke" received remainder and was executor.
